Selank is a synthetic heptapeptide (Thr-Lys-Pro-Arg-Pro-Gly-Pro) developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, based on the immune-modulating tetrapeptide Tuftsin with three amino acids added for stability. Approved as an anxiolytic in Russia since 2009, it demonstrated equivalent efficacy to medazepam (a benzodiazepine) in a randomised controlled trial with zero sedation, cognitive impairment, or withdrawal effects. It modulates over 800 genes in the rat hippocampus under stress conditions.

In vitro and preclinical mechanistic observations
Positively modulates GABA-A receptors through an allosteric mechanism similar to benzodiazepines, producing anxiolytic effects without downregulation or physical dependence.
Significantly increases Brain-Derived Neurotrophic Factor mRNA and protein in rat hippocampus and frontal cortex — effects persisting beyond the treatment period.
Inhibits enkephalin breakdown (endogenous opioid peptides), enhancing mood and stress tolerance without activating classical opioid receptors.
Upregulates IL-2 production and natural killer cell activity — inherited from its Tuftsin backbone — providing simultaneous anxiolytic and immune-supporting properties.
